Comparative study of three analysis methods (TTGE, flow cytometry and HPLC) for xenobiotic impact assessment on phytoplankton communities ArchiMer

The impacts of the fungicide Opus(A (R)) (epoxiconazole) on marine phytoplankton communities were assessed in a 12-day field experiment using in situ microcosms maintained underwater at 6 m depth. Three community analysis methods were compared for their sensitivity threshold in fungicide impact detection. When phytoplankton communities were exposed to 1 mu g l(-1) of epoxiconazole, no effects could be demonstrated using TTGE (Temporal Temperature Gradient gel Electrophoresis), flow cytometry or HPLC. When exposed to 10 mu g l(-1), TTGE fingerprints from PCR amplified 18S rDNA of communities exhibited significant differences compared with controls (ANOSIM, P = 0.028). In-situ microcosms, a tool for assessment of pesticide impacts on oyster spat (Crassostrea gigas) ArchiMer

Effects of the herbicide Basamais (bentazon) and the fungicide Opus (epoxiconazole) on oyster spat (Crassostrea gigas) were assessed using in-situ microcosms in a field experiment lasting 13 days. Six-week-old hatchery spat (mean size 1.1 mm), previously collected on PVC plates, was immersed in glass bottles filled with 200 mu m filtered seawater. Bottles were maintained underwater at 6 m depth and their water content changed every other day. Growth, measured as shell area index increase, was 126 +/- 4% in the control bottles. Resistance to triazole fungicides in Pyricularia species associated with invasive plants from wheat fields in Brazil Agronomy

Triazole fungicides have not been effective for managing the wheat blast disease in Brazil. A broad analysis across six geographical populations of Pyricularia graminis-tritici in central-southern Brazil indicated a high level of resistance to triazole fungicides. Since P. graminis-tritici is also associated with others poaceous species, here, we analyzed whether triazole-resistant isolates of the blast pathogen could be recovered from other poaceous hosts that are invasive of sprayed wheat fields. Toxicity of Epoxiconazole to the Marine Diatom Chaetoceros calcitrans: Influence of Growth Conditions and Algal Development Stage ArchiMer

The triazole fungicide epoxiconazole is extensively used to control fungi on crops and may present some potential risk from runoff on coastal ecosystems located close to agricultural areas. Phytotoxicity assessments were conducted on the marine diatom Chaetoceros calcitrans using both the active ingredient and its formulated product (Opus). The 3-day EC50 using cell count was 2.31 mg/L for epoxiconazole active ingredient and 2.9 mu g/L for epoxiconazole-formulated. The fungicide produced an increase of cellular volume, pigment (chlorophylls a, c, and carotenoids) content, ATP synthesis, and rates of photosynthesis and respiration.
